Restriction digests of the clone give the following sizes (kb): AccI--4.5; XhoI--uncut; HindIII--4.5; EcoRI--2.8, 1.75; KpnI--4.5. CYP2A (pMP12) and CYP2B (pMP17) hybridize to the same MluI (350 kb), NotI (520 kb), and SstII (350 kb) fragments, but to different BssHII, NaeI, NarI, SfiI, and SmaI fragments. This probe detects the SstI (SacI) RFLP originally reported by Wainwright. Three sizes of transcripts (1.9 kb, 2.7 kb, and 3.8 kb) are identified in Northern blots from human liver, with the 1.9 kb transcript being most prevalent. Each transcript demonstrated significant inter-individual variation (n=6). Enzyme(s) not detecting polymorphism: ApaI, BamHI, BglII, EcoRI, EcoRV, HaeIII, HindIII, HinfI, MspI, PstI, PvuII, RsaI, TaqI and XmnI. |
