Restriction digests of the clone give the following sizes (kb): BamHI--4.4, 1.8; EcoRI--6.4; BglII--6.4; HindIII--6.4.
The BamHI fragment has been used to construct high-level expression vectors for production of fusion proteins.
The additional glycine and serine residues at the carboxy terminus of the protein do not appear to affect amylase activity.
The vector was derived from pBD64 by replacing the PvuII site with a HindIII site.
Altered from the wild-type sequence by inserting a second BamHI site between the last codon and the stop codon.
The order of the major features in this plasmid is: HindIII - BamHI/amylase/BamHI - EcoRI - kanR/BglII - cmlR.
